Liquid Chromatography with LTQ Orbitrap mass spectrometer

The mass spectrometer is coupled to a UHPLC Ultimate 3000 (Dionex) for metabolomic analyzes. The ionization of the sample is of electrospray (ESI) type and can be done in positive and/or negative mode. The mass range can range from 0 to 2000 in m/z ratio, at the LTQ trap (first analyzer) and the orbitrap (second analyzer). This mass spectrometer also has a direct infusion device, offering maximum optimization of the detection conditions. We use Xcalibur (Thermo) software and Worflow4Metabolomics for data processing, spectrum analysis, statistical analysis, open database querying ... This workflow allows to obtain a file regrouping the masses eluted at the same retention time in the same group (pcgroup), any adducts and/or isotopes, to target a particular zone during an analysis in non-targeted mode. The platform has certain standards (amino acids, fatty acids, sterols ...) for targeted analysis. Coupled with the UHPLC, it is used for analyzes (qualitative and/or quantitative) of marine metabolites: seawater, fluids, tissues ... More specifically, the platform analyzes for example collodaries, bacteria, microalgae, dinoflagellates (eg: gymnodinium, brandtodinium and pelagodinium) ...
